Pharmacological Actions

Pharmacodynamics:

Doxycycline is a broad-spectrum tetracycline-class antibiotic with broad bacteriostatic activity. Susceptible organisms include Gram-positive bacteria such as Streptococcus pneumoniae, streptococci, certain staphylococci, Bacillus anthracis, Clostridium tetani, and Corynebacterium spp., as well as Gram-negative bacteria including Escherichia coli, Pasteurella spp., Salmonella spp., Brucella spp., Haemophilus spp., Klebsiella spp., and Burkholderia mallei. It also has moderate inhibitory activity against Rickettsia, Mycoplasma, and Spirochetes.

Pharmacokinetics:

Rapidly absorbed following oral administration with minimal food effect and high bioavailability. Maintains effective plasma concentrations over an extended period, with strong tissue penetration and wide distribution, including intracellular accumulation. The apparent volume of distribution at steady state in dogs is approximately 1.5 L/kg. Protein binding is high: 75-86% in dogs. A portion is inactivated in the intestine via chelation; approximately 75% of the administered dose is eliminated by this route in dogs. Renal excretion accounts for approximately 25%, and biliary excretion for less than 5%. The elimination half-life in dogs is approximately 10-12 hours.

 

Drug Interactions

(1) Concomitant administration with sodium bicarbonate raises gastric pH, reducing absorption and activity of this product.

(2) Divalent and trivalent cations form complexes with doxycycline. Concurrent administration with antacids containing calcium, magnesium, or aluminum, iron-containing preparations, or dairy products will reduce absorption and lower plasma drug concentrations.

(3) Concomitant use with potent diuretics such as furosemide may exacerbate renal impairment.

(4) May interfere with the bactericidal activity of penicillin-class antibiotics against actively dividing bacteria; concurrent use should be avoided.

 

Indications

Tetracycline-class antibiotic. For the treatment of infections caused by Gram-positive bacteria, Gram-negative bacteria, Mycoplasma, and other susceptible organisms.

 

Dosage and Administration

Expressed as doxycycline. Oral administration: 5-10 mg per 1 kg body weight per dose for dogs and cats, once daily for 3-5 consecutive days, or as directed by a veterinarian.

 

Adverse Reactions

(1) Vomiting may occasionally occur after oral administration. Esophagitis may develop if the tablet adheres to the esophageal mucosa.

(2) Disruption of intestinal microflora may occur, prolonged use may occasionally cause vitamin deficiency and, in severe cases, superinfection. Rare secondary infections with resistant Salmonella or unidentified pathogens and occasional severe diarrhea have been reported.

(3) Overdosage may cause gastrointestinal disturbances including anorexia, vomiting, or diarrhea.

 

Precautions

(1) Contraindicated in animals with severe hepatic or renal impairment.

(2) Avoid concurrent administration with dairy products or foods with high calcium content.

 

Withdrawal Period

Not required.
